Synonym | FGF2, BFGF, FGF-2, FGFB, HBGF-2, fibroblast growth factor 2 |
Background | FGF2, also known as basic fibroblast growth factor (bFGF) and FGF-beta, is a growth factor and signaling protein encoded by the FGF2 gene.It is synthesized primarily as a 155 amino acid polypeptide, resulting in an 18 kDa protein. However, there are four alternate start codons which provide N-terminal extensions of 41, 46, 55, or 133 amino acids, resulting in proteins of 22 kDa (196 aa total), 22.5 kDa (201 aa total), 24 kDa (210 aa total) and 34 kDa (288 aa total), respectively.Generally, the 155 aa/18 kDa low molecular weight (LMW) form is considered cytoplasmic and can be secreted from the cell, whereas the high molecular weight (HMW) forms are directed to the cell's nucleus. |
Description | A DNA sequence encoding the mature form of human bFGF (Pro 143-Ser 288) was expressed, with an additional Met at the N-terminus. |
Source | E.coli |
Predicted N Terminal | Met |
AA Sequence | Pro 143-Ser 288 |
Molecular Weight | The recombinant human bFGF is a disulfide-linked homodimeric protein. The reduced monomer consists of 147 amino acids and has a calculated molecular mass of 16.5 kDa. In SDS-PAGE under reducing conditions, the apparent molecular mass of bFGF is approximately 17 kDa. |
Purity | >95% as determined by SDS-PAGE |
Endotoxin | < 1.0 EU per mg protein as determined by the LAL method |
Bioactivity | Measured in a cell proliferation assay using BALB/c 3T3 mouse embryonic fibroblasts. The ED50 for this effect is typically 0.02-0.1 ng/ml. |
Formulation | Recombinant Human FGF2 Protein was lyophilized from sterile 20mM Tris, pH 7.4 |
Stability | Recombinant Human FGF2 protein is stable up to 1 year at -70°C |
Usage | For Research Use Only |
Storage | Store the protein under sterile conditions at -20°C to -80°C. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles. |
