Product Overview
Description | GPRC5D human recombinant protein generated in vitro E.coli expression system is a full-length polypeptide chain containing 1-350aa and having a molecular weight of about 40 kDa on SDS-PAGE. GPRC5D is expressed with a 10xHis-tag at C-terminus. Its purity is greater than 90% measured by SDS-PAGE. GPRC5D is a 7-pass transmembrane receptor protein primarily expressed in plasma-cell phenotypic cells, including the majority of malignant plasma cells from multiple myeloma (MM) patients. It is also actively expressed in hair cells, lung tissue, and bone marrow. The high expression on MM cells makes GPRC5D become an attractive target for MM's treatment. Anti-GPRC5D CAR-T cells provide an alternative therapy in case of recurrence after anti-BCMA therapy since GPRC5D expression is independent of BCMA expression. BCMA binding contributes to MM cells' proliferation and survival. |

Form | Liquid or Lyophilized powder |
Buffer | Liquid form: default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. Lyophilized powder form: the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0. |
Reconstitution | Briefly centrifuged the vial prior to opening to bring the contents to the bottom. Reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. It is recommended to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. The default final concentration of glycerol is 50%. |
Storage | 1. Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. 2. Avoid repeated freeze-thaw cycles. 3. Store working aliquots at 4°C for up to one week. 4. In general, protein in liquid form is stable for up to 6 months at -20°C/-80°C. Protein in lyophilized powder form is stable for up to 12 months at -20°C/-80°C. |
Notes | Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week. |
Target Function | Receptor for the chemotactic and inflammatory peptide anaphylatoxin C5a. The ligand interacts with at least two sites on the receptor: a high-affinity site on the extracellular N-terminus, and a second site in the transmembrane region which activates downstream signaling events. Receptor activation stimulates chemotaxis, granule enzyme release, intracellular calcium release and superoxide anion production. |
Subcellular Location | Cell membrane; Multi-pass membrane protein. Cytoplasmic vesicle. |
Protein Families | G-protein coupled receptor 1 family |
