Product Overview
Tag | His&Fc |
Host Species | Human |
Accession | NP_001036064.1 |
Synonym | ERBB4, ALS19, HER4, p180erbB4, erb-b2 receptor tyrosine kinase 4 |
Background | Receptor tyrosine-protein kinase erbB-4 is an enzyme that in humans is encoded by the ERBB4 gene. Alternatively spliced variants that encode different protein isoforms have been described; however, not all variants have been fully characterized. Receptor tyrosine-protein kinase erbB-4 is a receptor tyrosine kinase that is a member of the epidermal growth factor receptor family. ERBB4 is a single-pass type I transmembrane protein with multiple furin-like cysteine rich domains, a tyrosine kinase domain, a phosphotidylinositol-3 kinase binding site and a PDZ domain binding motif. The protein binds to and is activated by neuregulins-2 and -3, heparin-binding EGF-like growth factor and betacellulin. Ligand binding induces a variety of cellular responses including mitogenesis and differentiation. Multiple proteolytic events allow for the release of a cytoplasmic fragment and an extracellular fragment. |
Description | A DNA sequence encoding the extracellular domain (Met 1-Pro 651) of human ErbB4 precursor, expressed in HEK293 cells was fused with the C-terminal His-tagged Fc region of human IgG1 at the C-terminus. |
Source | HEK293 |
Predicted N Terminal | Glu 26 |
AA Sequence | Met 1-Pro 651 |
Molecular Weight | The recombinant human ErbB4/Fc is a disulfide-linked homodimeric protein after proteolytic removal of the signal peptide. The reduced monomer consists of 871 amino acids and predicts a molecular mass of 97.5 kDa. As a result of glycosylation, the rh ErbB4/Fc monomer migrates as approximately 125-135 kDa band in SDS-PAGE under reducing conditions. |
Purity | >95% as determined by SDS-PAGE |
Endotoxin | < 1.0 EU per μg of the protein as determined by the LAL method |
Bioactivity | 1. Measured by its ability to bind biotinylated human Fc-NRG1 (isoform Beta 1) in a functional ELISA.2. Measured by its ability to bind with biotinylated human NRG1 (isoform Beta 1) in a functional ELISA.3. Measured by its ability to bind with biotinylated human NRG1 (aa 2-246)-Fc in a functional ELISA.4. Measured by its ability to bind with biotinylated human NRG1 (aa 20-241)-His in a functional ELISA. |
